Synthetic Nanodisc全長多穿膜蛋白

Nanodisc中文名稱為納米圓盤,該平臺利用具有疏水和親水雙重特性的物質(zhì)作為穩(wěn)定劑,穩(wěn)定劑朝向內(nèi)部脂層的疏水面可將膜蛋白整合到Nanodisc中,維持膜蛋白的天然空間構(gòu)象和活性。同時(shí),朝外的親水面使得Nanodisc在水溶液中具有很高的溶解度和穩(wěn)定性。
與市面上大多數(shù)MSP Nanodisc不同,締碼生物研發(fā)的Synthetic Nanodisc能直接從完整的細(xì)胞中制作。在這個(gè)過程中,使用的合成高分子具有雙重功能。首先,它溶解細(xì)胞膜,類似于洗滌劑,同時(shí)利用天然細(xì)胞磷脂在膜蛋白周圍形成納米盤結(jié)構(gòu)。此外,締碼制備Nanodisc蛋白時(shí)使用的是哺乳動(dòng)物表達(dá)系統(tǒng),這可確保蛋白在結(jié)構(gòu)、功能和翻譯后修飾方面與天然對應(yīng)物非常相似,使其成為生物化學(xué)和生物醫(yī)學(xué)領(lǐng)域廣泛應(yīng)用的理想工具。
